摘    要:目的:建立一年蓬中焦袂康酸的HPLC含量测定方法,并对一年蓬药材不同部位中焦袂康酸含量进行测定。方法:采用Unitary C18(250 mm×4.6 mm,5μm)色谱柱,以甲醇-1‰三氟乙酸水溶液(5∶95)为流动相,检测波长为270 nm,柱温为30℃,流速为1.0 mL·min^-1。结果:焦袂康酸的线性范围为0.25~62.0μg·mL^-1(r=0.999 3),平均加样回收率为98.4%,RSD=1.2%(n=6)。一年蓬花、叶、茎中焦袂康酸含量分别为2.56,0.40,0.08 mg·g^-1,根中含量低于0.012 mg·g^-1。结论:该方法简便、准确、重复性好,可以作为一年蓬中焦袂康酸的含量测定方法,为合理利用一年蓬药材提供参考依据。

Abstract:Objective: To establish a method for the determination of pyromeconic acid in Erigeron annus by HPLC,and determine the contents of pyromeconic acid in different parts of E. annus. Methods:Unitary C18 column(4. 6 mm × 250 mm,5 μm) was used with a mobile phase consisting of methanol- 1‰ trifluoroacetic acid aqueous(5∶95). The detection wavelength was 270 nm. Column temperature was 30 ℃. The flow rate was 1. 0 mL·min^- 1. Results: The linear range of pyromeconic acid was 0. 25 ~ 62. 0 μg·mL^- 1(r = 0.999 3)and The mean recoveries rate was 98.4 %,RSD 1.2%(n = 6). The contents of Pyromeconic acid in flower,leaf,stem were 2. 56,0. 40,0. 08 mg·g^- 1,while root was lower than 0.012mg· g^- 1. Conclusion: The method is simple, accurate, reproducible, and can be used as the determination method of pyromeconic acid in E. annus,and meanwhile,can provide reference basis for the rational use of E. annus.
