摘 要: | 目的:研究回生口服液调控Wnt信号传导通路细胞周期蛋白D1(CyclinD1)影响人肺腺癌细胞(LAC)增殖的机制。方法:制备高、中、低剂量(11.6g/kg,5.8g/kg,2.9g/kg)回生口服液含药血清,与人肺腺癌细胞系共同培养,MTT法检测含药血清对肺腺癌细胞的抑制率,流式细胞仪检测肺腺癌细胞周期相对分布;建立C57BL/6人肺腺癌荷瘤小鼠模型,予回生口服液高、中、低剂量治疗,免疫组化法检测小鼠移植瘤肺腺癌细胞Cyclin—D1阳性表达水平。结果:与阴性对照组比较,回生口服液高、中剂量组及顺铂组含药血清对肺腺癌细胞的生长抑制率明显增高,且干预的肺腺癌细胞G1期细胞比例明显增高,s期、G2期细胞比例明显减少(P〈0.01,P〈0.05);回生口服液中、高剂量及顺铂(DDP)治疗组小鼠肺腺癌细胞内CyclinD1阳性表达率明显低于模型组(P〈0.01)。结论:回生口服液可明显抑制人肺腺癌细胞生长,使癌细胞停滞于细胞周期的G1期,并可明显降低肺腺癌细胞内CyclinD1的阳性表达。

Abstract: | Objective: To study the mechanism of Hui-sheng oral liquid affecting the proliferation of the human lung adenocar- cinoma cells ( LAC ) through controlling CyclinD1 of Wnt signal pathway. Methods : Drug serum of high, middle and low-close Hui- sheng oral liquid ( 11.6 g/kg, 5.8 g/kg, 2. 9 g/kg) were prepared. The inhibitory rate of LAC cells was calculated by MTT, and the cell cycle was analyzed by the flow cytometry. C57BL/6 solid tumor model mice were established, treated by high, middle and low-dose Hui-sheng oral liquid ( 11.6g/kg, 5.8g/kg, 2.9g/kg) for 14 days, then their tumors were taken out and the expression of CyclinD1 were detected by the immunohistochemical assay. Results: The inhibitory rate of LAC cells intervened by middle, high-dose Hui-sheng oral liquid and DDP were apparently higher than the negative control group, cells in G1 phase increased, while cells in G2 phase and S decreased (P 〈0. 01, P 〈0.05) ; The middle, high-dose Hui-sheng oral liquid and DDP could regulate down the ex- pression of CyclinD1 in the tumor (P 〈 0. 01 ) . Conclusion: Hui-sheng oral liquid could decrease the expression of CyclinD1 of LAC in Wnt Signal Pathway and prevent their G1 period turning into S period, therefore inhibit the growth of lung cancer cells. |
