基于热敏通道TRPV1探讨吴茱萸汤对醋酸内脏痛模型小鼠的止痛作用

摘    要:目的:探讨吴茱萸汤对醋酸诱导的内脏痛模型小鼠的止痛作用。方法:在醋酸诱导的内脏痛模型小鼠上,结合扭体实验及免疫组织化学染色探查吴茱萸汤的止痛作用与TRPV1的关联。结果:与模型组对比,吴茱萸汤组可显著降低小鼠扭体行为差异极显著(P0.01),同时,吴茱萸汤组小鼠第1次扭体行为出现时间明显延迟,差异显著(P0.05);与正常组对比,模型组结肠组织中TRPV1阳性表达显著增多差异极显著(P0.001),而吴茱萸汤组小鼠结肠粘膜层与肌层的TRPV1阳性表达明显减少,与模型组对比差异极显著(P0.001)。结论:热敏通道TRPV1是吴茱萸汤发挥止痛作用的关键环节。

Abstract:Objective:To investigate the analgesic effect of Wuzhuyu tang on acetic acid-induced visceral pain in mice.Methods:Animal behavior experiment and immunohistochemical technique were used to observe the analgesic effect of Wuzhuyu Tang and the expression of TRPV1 in colon tissue of acetic acid-induced visceral pain mice.Results:The frequency of writhing behavior in Wuzhuyu tang group was significantly lower than that in model group (P < 0.01).Wuzhuyu tang was able to delay the occurrence time of writhing behavior in mice,and the difference was significant (P < 0.05) compared with model group.The expression of TRPV1 in colonic tissue of model group was significantly higher than that of normal group (P < 0.001).After 4d administration of Wuzhuyu tang,the expression of TRPV1 in colonic tissue was significantly lower than that in model group (P < 0.001).Conclusion:Thermosensory channel TRPV1 play a key role on the analgesic effect of Wuzhuyu tang.
