正交试验优选苦参方中苦参有效成分的提取工艺

摘    要:目的:优选中医治疗过敏性皮肤病的传统方剂苦参方中苦参有效成分的最佳提取工艺。方法:以苦参主要有效成分苦参碱和氧化苦参碱的含量及浸膏得率为考察指标,选择乙醇浓度、乙醇用量、提取时间为考察因素,采用L_9(3~4)正交试验法确定最佳提取工艺。结果:乙醇浓度和提取时间对提取效率有显著性影响(P<0.05),乙醇用量的影响无显著性意义(P>0.05),最佳提取工艺为8倍量于药材的60%乙醇回流提取2次,每次3 h。结论:优选确定的工艺可用于苦参方中苦参有效成分的提取。

Abstract:Objective:To optimize the technology for extraction of active principles of Radix Sophorae Flavescentis in Kushen recipe which has been used for treating allergic skin diseases in China for a long time.Methods:L_9 (3~4) orthogonal test was used to optimize the technology for extracting matrine and oxymatrine,with the contents of matrine and oxymatrine and the yield of extract serving as the criteria.The influential factors such as concentration and volume of ethanol,time of extrac- tion were evaluated,and each factor had three levels.Results:The concentration of ethanol and the time of extraction had sig- nificant influence on the extraction efficiency.The optimal extraction technology was as follows:the medicinal material was refluxed with 60% ethanol aqueous solution for two times and each time was 3 hours.Conclusion:The optimized extraction technology is practical.
