摘    要:目的:探讨西施口服液延缓小鼠皮肤衰老的药理作用。方法:将50只小鼠随机分为正常对照组、衰老模型组和西施口服液低、中、高剂量组(2.0,4.0,8.0 g·kg-1)。除正常对照组外,其余各组采用5% D-半乳糖颈背皮下注射建立皮肤衰老小鼠模型,造模成功后进行给药处理。治疗结束后,检测小鼠皮肤细胞病理形态、表皮和真皮的厚度、含水量和皮肤组织匀浆中SOD、GSH-PX活力、Hyp、LPO及·O-2、·OH、H2O2等自由基浓度的变化。结果:与正常对照组相比,模型组小鼠皮肤表皮和真皮明显变薄,细胞形态不规则,胶原纤维束变细且多处断裂,羟脯氨酸(Hyp)含量降低;生化检测表明,皮肤组织中SOD、GSH-PX活力均比正常组减少(P均<0.01),而LPO、·O-2、·OH、H2O2等自由基浓度均大幅高于正常组(P均<0.01)。经药物治疗30天后,西施口服液低、中、高3个组小鼠皮肤中的SOD、GSH-PX活力均明显升高(P<0.01或P<0.05);Hyp、LPO及·O-2、·OH、H2O2等自由基浓度均明显减少(P<0.01或P<0.05),与此同时,实验各组小鼠皮肤厚度、含水量均明显上升(P<0.01或P<0.05),与模型组相比,差异极显著。结论:西施口服液可明显改善衰老小鼠皮肤的病理变化,增加皮肤的弹性和与质量,显著提高衰老小鼠皮肤中抗氧化酶的活力,降低脂质过氧化物和自由基的含量;表明本品有较好的抗皮肤衰老的药理作用。

Abstract:OBJECTIVE To explore pharmacological effects of Xishi oral liquid on skin aging mice.METHODS Fifty mice were randomly divided into five groups:normal control group,aging model group,treatment groups with low,middle and high doses of Xishi oral liquid (2.0,4.0,8.0 g·kg-1).Mouse model of skin aging was established by subcutaneous injection of 5% D-galactose (D-gal) into nape skin except for mice in normal control group.Then mice of treatment groups were treated with Xishi oral liquid for 30 days.After treatment,skin morphological structure,skin thickness and water content were observed.Activities of SOD and GSH-PX,concentrations of Hyp and LPO,and free radicals such as ·O-2,·OH and H2O2 were detected.RESULTS Compared with control group,aging model group showed that thicknesses of dermis and epidermis were significantly reduced with lower Hyp concentration.Most cells had irregular shapes,and collagen fibers were thinner and broken.Activities of SOD and GSH-PX were statistically decreased,whereas contents of ·O-2,·OH and H2O2 were remarkably increased (P<0.01).After treatment for 30 days,activities of SOD and GSH-PX of three treatment groups showed obvious increases,while Hyp,LPO and free radicals showed significant decreases (P<0.01 or P<0.05).Skin thickness and water content raised significantly compared with model group (P<0.01 or P<0.05).CONCLUSION Xishi oral liquid can improve pathological morphology of skin aging mice,thus increase elasticity and quality of skin.Meanwhile,it also increases activities of antioxidases and decreases lipid peroxide and free radicals.In conclusion,Xishi oral liquid shows pharmacological effects of anti-aging on skin.
