| 摘 要： | 目的研究葫芦素E(cucurbitacin E,CuE)体外对人肝癌BEL7402和HepG2细胞的增殖抑制作用及机制。方法MTT法检测不同浓度CuE体外对BEL7402和HepG2细胞的增殖抑制作用;流式细胞仪检测CuE对细胞周期分布的影响;Western blot法检测CuE对细胞周期调节蛋白cy-clinB1、Cdc25C、Cdk1及p21表达的影响。结果CuE作用72 h可剂量依赖性地抑制BEL7402和HepG2细胞增殖;CuE(100 nmo.lL-1)作用12、24 h后可使BEL7402和HepG2细胞周期阻滞于G2/M期,并可下调Cdk1蛋白的表达,上调p21蛋白的表达。结论CuE体外可抑制人肝癌BEL7402和HepG2细胞的增殖,其增殖抑制作用与诱导G2/M周期阻滞有关。
