摘    要:目的:研究尿激酶型纤溶酶原激活物(uPA)对高糖诱导系膜细胞(MC)增殖及α-肌动蛋白(α-SMA)表达的影响.方法:MC分组:对照组(5 mmol/L D-葡萄糖),高糖组(30 mmol/L D-葡萄糖)和uPA组(30 mmol/L D-葡萄糖+不同浓度uPA).分别用四甲基偶氮唑盐比色法(MTT法)和流式细胞仪检测细胞增殖和细胞周期情况;激光共聚焦观察α-SMA表达.结果:高糖刺激MC增殖率明显增加,uPA呈剂量依赖性的阻止细胞由G0/G1期进入S期,抑制MC增殖.高糖增加α-SMA表达,随着uPA浓度增加,核周α-SMA表达较高糖组明显减少(P<0.01).结论:uPA抑制高糖诱导的系膜细胞增殖和表型转化,对糖尿病肾小球硬化有一定的治疗作用.

Urokinase-Type Plasminogen Activator Inhibits the Proliferation and Alpha-Smooth Muscle Actin Expression of Glomerular Mesangial Cell
Abstract:Objective: To evaluate the effect of urokinase-type plasminogen activator(uPA) on high-glucose induced-proliferation and alteration of α-SMA expression of glomerular mesangial cell(MC).Methods: MCs were exposed to high-glucose(30 mmol/L D-glucose) medium or high glucose plus different concentration of uPA.The cell cycle and proliferation were respectively assessed by flow cytometry and MTT assay.Laser scanning confocal microscope was used to observe the expression of α-SMA.Results: uPA prevented high glucose-exposed MC from G0/G1 phase into S phase in a dose-dependent manner,thus inhibited proliferation of MC.High glucose exposure increased perinuclear expression of α-SMA in MC,which was prevented by uPA in a dose-dependant manner.Conclusion: uPA may exert a therapeutic effect on diabetic glomerulosclerosis through inhibiting the proliferation and phenotypic change of MC.
