心钠素在肿瘤细胞增殖和凋亡中的作用

目的研究心钠素(ANP)诱导体内和体外肿瘤细胞增殖和凋亡的作用,探索ANP抗肿瘤的作用机制。方法通过体外细胞培养,用CyQUANT试剂盒和APO-BRDU试剂盒分别检测不同浓度的ANP对黑色素瘤细胞增殖和凋亡的影响。通过微量泵将ANP输入带瘤小鼠,14 d后取出肿瘤,用Ki67抗体和Tunel试剂分别检测肿瘤切片中黑色素瘤细胞的增殖和凋亡。结果ANP能促进小鼠体内黑色素瘤细胞的凋亡,凋亡指数是对照组的3倍(P0.01),但ANP不能影响黑色素瘤细胞的增殖。ANP在体外细胞培养中既不能调节小鼠黑色素瘤细胞的增殖,也不能调节其凋亡。结论ANP可能部分地通过促进体内肿瘤细胞凋亡从而抑制肿瘤的生长,此结果有助于进一步探讨ANP抗肿瘤的作用机制。

Effects of atrial natriuretic factor on tumor cell proliferation and apoptosis

Objective It is to study the effects of atrial natriuretic factor(ANP) on melanoma cell proliferation and apoptosis in vivo and in vitro and explore the mechanism of ANP inhibiting the growth of tumor.Methods Melanoma cells were cultured with ANP in a variety of concentrations in vitro.CyQUANT cell proliferation assay kit and APO-BRDU in situ cell death detection kit were used to detect cellular proliferation and apoptosis.ANP was infused into tumor-bearing mice by micro-pumps.Tumors were removed 14 days later.Proliferated and apoptotic melanoma cell in tumor slices were detected by Ki67 antibody and Tunel reagents.Results ANP could promote the apoptosis of melanoma cells in vivo of mice.The apoptotic index of melanoma cells in ANP group was three times at control group(P<0.01).ANP could neither regulate the proliferation of melanoma cells in vitro of mice,nor regulate the apoptosis.Conclusion ANP may inhibit tumor growth through partly promoting the apoptosis of tumor cells in vivo.The result will be useful for further approaching the mechanisms of ANP inhibiting tumor growth.
