喉部鳞状细胞癌组织中p_(53)蛋白的免疫转印分析

摘    要:为观察喉部鳞状细胞癌及其癌旁组织中P53蛋白的降解及其寡聚物的形成,采用免疫转印法分析了15例喉部鳞状细胞癌标本中P53蛋白的电泳行为。发现P53蛋白阳性率在癌灶中心组织中为100%,在癌旁组织中为66.7%,两组间相差显著(P<0.05)。在所有标本均未发现P53蛋白寡聚物,且P53蛋白均有不同程度的降解,其中发生完全降解者(分子量均低于53kD)在癌灶中心组织中为60.0%,在癌旁组织中为30.0%(占阳性例数的比例),且癌旁组织中P53蛋白的平均分子量较癌灶中心组织低,但是,无统计学意义(P>0.05)。

Abstract:In order to investigate the hydrolysis or oligopolymerization of p53 protein in the tissues of laryngeal squamous cell carcinoma, wer have observed their electrophoretic action in 15 cases of the disease with Western blotting method. We found that the positive rate of p53 protein in the center carcinous tissues (100% ) was significantly higher than in the tissues adjacent to cancer (66. 7% ), (P<0. 05). No oligo-polymerization of p.3 protein was found in all the tissues, but hydrolyzed to a greater or lesser degree. Wholly hydrolytic (molecular weight of any bands below 53kD) rate was 60% in the center carcinous tissues and 30% in the adjacent tissues (of positive cases) respectively. The average molecular weight of p53 protein in the adjacent tissues was lower than in the center carcinous tissues, but not significantly (P>0.05). We speculate that during the canceration of laryngeal squamous cell carcinoma, mutated p53 protein mainly plays its cancerigenic role through acquire new function of overthrowing the negative regulating role of wild-type but not oligo-polymerization. Wild-type p53 protein may be hydorlyzed and lost negative regulating function. Mutated p53 protein was overexpressed in center carcinous tissues.
