星点设计 - 效应面优化法优化龙胆苦苷的纯化工艺

摘    要: 目的 采用星点设计 - 效应面优化法研究 HPD300 型大孔吸附树脂分离纯化龙胆中龙胆苦苷的工艺条件及参数。 方法 ■ 以上柱液中龙胆苦苷的浓度、洗脱液中乙醇的浓度和洗脱液用量为考察因素,以产物中龙胆苦苷转移率和百分含量为指标,采用多元线性回归及二次多项式拟合,建立指标和考察因素之间的数学关系式,根据最佳数学模型描绘效应面,再根据效应面优选最佳纯化工艺。 结果 二次多项式能够较好的描述指标与因素之间的关系。最佳纯化工艺龙胆苦苷转移率和百分含量理论预测值与实测值偏差较小。 HPD300 树脂分离纯化龙胆苦苷的纯度可达 78.5 %,说明优化得到的纯化工艺能够有效的分离龙胆中水溶性的裂环环烯醚萜苷类成分。 结论 星点设计 - 效应面优化法可用于龙胆苦苷纯化工艺的优化,模型预测性良好,实验设计有较高的可靠性。

Optimized Purification Process of Gentiopicrin by a Central Composite Design-Response Surface Methodology

Abstract:OBJECTIVE To study the technological parameters of the purification process of gentiopicrin in Gentiana scabra Bge. with HPD-300 type macroreticular resin by a central composite design-response surface methodology (CCD-RSM). METHODS Independent variables were sample concentration, eluent concentration and eluent volume, while dependent variables were transferring rate and content of gentiopicrin in product. Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables. The response surface and overlay contour plot were delineated according to best-fit mathematic model, and optimum technological parameters were selected by the overlay contour plot. Prediction was carried out through comparing the observed and predicted values. RESULTS Two order quadratic equation described the relationship between dependent and independent variables, with a regression coefficient of 0.946 9 and 0.973 4. Bias between observed and predicted values of transferring rate and content of gentiopicrin were -1.26% and -0.33%. The content of gentiopicrin in the final product was up to 78.5%, while it was just 7.53% before purification. The optimized purification process was suitable for the separation of water-soluble secoiridoid glycoside from Gentiana scabra Bge.. CONCLUSION CCD-RSM can be used to optimize the purification parameters of gentiopicrin, and the optimum mathematic model is highly predictive.
