Tspan-5在高转移大肠癌细胞系中的表达及其对黏附行为影响的研究

摘    要:目的:研究四分子交联体5(tetraspanin5,Tspaw5)在不同转移潜能大肠癌细胞株中的差异表达及其与大肠癌细胞异质黏附能力的关系。方法:用Western—Blotting研究Tspan-5在3种细胞株的差异表达,用RNAi技术下调Tspan-5在高转移潜能LoVo细胞株中的表达,观察其对LoVo细胞在不同基底膜主要成分上黏附行为的影响。结果:Tspan-5在不同转移潜能大肠癌细胞株中存在差异表达,转移潜能最高的LoVo细胞株表达最强,转移潜能最低的LST—R1细胞株表达最弱,下调Tspan-5可显著抑制LoVo细胞在CollageonⅣ、Fibronectin、Laminin、Vitronectin上的黏附行为。结论:Tspan-5在高转移潜能的大肠癌细胞中过表达,并可调节大肠癌细胞的基底膜黏附力,其与大肠癌转移的关系值得进一步研究。

Abstract:Objective:To investigate different expression of Tspan-5 among three colorectal cancer (CRC) cell lines, and the relationship between colorectal cancer cell adhesion ability and Tspan-5 expression. Methods:Different expression of Tspan-5 among three cell lines were detected via Western-Blotting assays. RNAi technology was used to knock down Tspan-5 expression in LoVo cells. Adhesion ability was tested by adhesion experiment. Results:Differential expression of Tspan-5 among three cell lines was observed via Western-Blotting. Downregulation of Tspan-5 expression level could inhibit LoVo adhesion ability on a range of extracellular matrix components, suggesting that Tspan-5 mediates vital cellular functions. Conclusion:Tspan-5 was overexpressed in the highly metastatic CRC cell lines, and required for colorectal cancer cell adhesion. Tspan-5 might contribute to colorectal cancer progression and it is worth for further exploring.
