转化生长因子-β对大鼠腹膜纤维化的防治研究

摘    要:目的探讨腹膜纤维化的防治措施。方法Wistar大鼠30只随机分成三组。A组为模型组,每日腹腔注射4.25%的百特透析液20ml,B组为给药组每日腹腔注射透析液同时灌胃给药(血管紧张素转换酶抑制剂)一平苏,C组为空白对照组,每日腹腔注入生理盐水20ml。试验开始30天后取大鼠腹膜作病理组织学检查,同时收集血清和腹透液测定转化生长因子-β。结果A组和B组的腹膜明显纤维化并且转化生长因子-β血清和腹透液中都有增加,与C组差异都有显著性。结论高糖可以诱导腹膜纤维化。腹膜纤维化时转化生长因子-β血清和腹透液中都升高并且可被一平苏抑制。3平苏有抗腹膜纤维化的作用。

Abstract:Objective To investigate the prevention and treatment of peritoneal fibrosis in long-term peritoneal dialysis patients. Methods Thirty Wistar rats were divided into three groups,A group model group, received daily intraperitoneal injection of 20 mL 4.25% ,B medicine group, received daily intraperitoneal injection of 20 mL 4.25% and angiotensin-converting enzyme inhibitor (inhibace) 10mg/kg · d was given orally to rats by gastric gavage in the morning once a day C group control group received daily intraperitoneal injection of 20 mL 0. 85 % natrli cloride. Thirty days late TGF-β expression were examined in abdominal cavity liquid and serum of peritoneal dialysis rats . Histopathology of parietal peritoneum was examined. Results TGF-β obviously increased in Both A and B groups as compared to C group (P〈0. 05)so do histopathology of peritoneum fibrosis. Conclusions high glucose can induce peritoneal fibrosis the expression of TGF- βboth in abdominal cavity liquid and serum is up-regulated which is involved in the mechanism of peritoneal fibrosis, that can be inhibated by angiotensin-converting enzyme inhibitor inhibace). Inh/bace can relieve peritoneal fibrosis.
