Abstract:The effect of captopril (Cap) on angiontensinⅡ(AngⅡ)-increased Ca2+ in rabbit aortic smooth muscle cells (ASMC) was examined. Cultured ASMC were loaded with the fluorescent dye Fura 2-AM and cellular free calcium ([Ca2+i) was determined by using digital image processing technique. AngⅡ induced a concentration-dependent increase in Ca2+i. Verapamil (0.1-10 μmol·L-1) inhibited AngⅡ-increased [Ca2+i, Cap 1-10 but not 0.1 μmol·L-1 decreased [Ca2+i response to AngⅡ. These results indicate that Cap inhibits Ang II-induced increase in [Ca2+i via voltage-dependent calcium channel, which may be of therapeutic relevance.
