Inhibition of proliferation and invasion of pancreatic cancer cells with AMD3100 in vitro

Objective To explore the effect of CXCR4 inhibitor AMD3100 on proliferation and invasion of pancreatic cancer cells and the its possible mechanism.Methods Human pancreatic cancer cells line SW1990 was treated with AMD3100 at different final concentrations,and then CCK-8 assay was used to detect the proliferation of SW1990.The transwell system pretreated with Matrigel was used to investigate the invasion of SW1990 treated with AMD3100.RT-PCR was used to detect the mRNA expressions of vascular endothelial growth factor(VEGF) and matrix metalloproteinase-9(MMP-9) in SW1990 cells treated with AMD3100.Results The proliferation of SW1990 was significantly surpressed by AMD3100 in a dose-dependent manner,especially when SDF-1 was added in.Aslo,AMD3100 could inhibit the invasion and migration of SW1990.The mRNA expressions of VEGF and MMP-9 reduced markedly in SW1990 treated with AMD3100.Conclusion AMD3100 inhibits proliferation and invasion activity of pancreatic cancer cells through down-regulation of VEGF and MMP-9 mRNA expressions.
