| 摘 要： | 目的 探讨利用绿色荧光蛋白(GFP)作为骨髓间充质干细胞(BMSCs)移植治疗脑出血示踪标记物的可行性.方法 利用GFP空载体重组腺病毒感染BMSCs,应用脑立体定位仪将感染的BMSCs移植入脑出血大鼠纹状体内,移植后5、14 d,脑组织行冰冻切片,荧光显微镜下观察BMSCs在脑组织内分布的表达.结果 感染细胞与未感染细胞体外培养形态无差异,感染细胞及其传代细胞发强绿色荧光.荧光显微镜下观察脑出血模型冰冻切片,移植后各时间点在移植部位周围病灶区内有大量发绿色荧光的细胞.结论 GFP感染BMSCs能在脑出血大鼠脑内稳定持久表达,表明GFP是脑出血干细胞移植治疗的有效示踪标记物,为进一步研究移植细胞在宿主脑内的可塑性研究奠定基础.
