溪黄草对circ_0091579/miR-515-5p、肺癌细胞A549增殖及凋亡的影响

摘    要:目的探讨溪黄草对circ_0091579/miR-515-5p及肺癌细胞A549增殖和凋亡的影响。 方法将肺癌A549细胞分为对照组、溪黄草(20、40、80 mg/L)组、si-circ_0091579组、si-NC组、pcDNA-circ_0091579组、pcDNA组、溪黄草+pcDNA-circ_0091579组。CCK-8、平板克隆实验、流式细胞术检测A549细胞抑制率、克隆形成数以及凋亡率。qRT-PCR检测circ_0091579和miR-515-5p表达。双荧光素酶报告实验验证circ_0091579和miR-515-5p相互作用。 结果溪黄草显著降低A549细胞的增殖和克隆形成能力,促进细胞凋亡,诱导cleaved-Caspase-3表达上调及circ_0091579下调,且呈质量浓度依赖性。下调circ_0091579可降低A549细胞的增殖和克隆形成能力,促进细胞凋亡,诱导cleaved-Caspase-3表达上调。上调circ_0091579可逆转溪黄草对A549细胞的增殖抑制和凋亡促进功能。circ_0091579和miR-515-5p直接特异性结合。 结论溪黄草可抑制肺癌细胞A549增殖,诱导其凋亡,其机制与下调circ_0091579/miR-515-5p途径有关。

Abstract:AimTo investigate the effect of Rabdosia lophanthoides on circ_0091579/miR-515-5p and proliferation and apoptosis of lung cancer cell A549. MethodsLung cancer A549 cells were divided into control group, Rabdosia lophanthoides (20,40, 80 mg/L) group, si-circ_0091579 group, si-NC group, pcDNA -circ_0091579 group, pcDNA group, Rabdosia lophanthoides+pcDNA-circ_0091579 group. CCK-8, plate cloning experiment and flow cytometry were performed to evaluate the inhibition rate, clone formation number and apoptosis rate of A549 cells. qRT-PCR was used to detect circ_0091579 and miR-515-5p expression. The interaction between circ_0091579 and miR-515-5p was verified via dual-luciferase reporter assays. ResultsRabdosia lophanthoides significantly reduced the proliferation and clonogenicity of A549 cells, promoted cell apoptosis, induced up-regulation of cleaved-Caspase-3 expression and decreased circ_0091579 level in a mass concentration dependent manner. Down-regulation of circ_0091579 reduced the proliferation and clonogenicity of A549 cells, promoted cell apoptosis, and induced up-regulation of cleaved-Caspase-3 expression. Up-regulation of circ_0091579 could reverse the proliferation inhibition and apoptosis promotion of A549 cells. circ_0091579 directly and specifically bound to miR-515-5p. ConclusionRabdosia lophanthoides inhibited the proliferation and induced apoptosis of lung cancer cell A549, which was associated with the down-regulation of circ_0091579/miR-515-5p pathway.
