宫颈尖锐湿疣组织中HPV分型与端粒酶活性的研究

摘    要:目的由于宫颈尖锐湿疣(CA)皮损中HPV高危型感染常与其癌变有关,而端粒酶活性增高多见于恶性疾病。该研究探讨宫颈CA皮损中HPV分型与端粒酶活性的关系。方法采用荧光定量PCR检测HPV病毒分型,端粒重复序列扩增文件-酶标法(TRAP—ELISA)检测端粒酶活性。结果尖锐湿疣患者皮损中端粒酶活性明显高于正常皮肤组织;24例(40.0%)HPV16/18型阳性,34例(56.7%)HPV6/11型阳性,2例未检测到HPV;HPV16/18型感染的皮损中端粒酶活性明显高于HPV6/11型感染者。结论认为端粒酶活性与HPV型别有关,可能与HPV病毒协同参与CA发病,HPV16/18型阳性者在临床可能有癌变倾向,抑制端粒酶活性可能可以阻碍疣体生长,为临床治疗提供一个新的方向。

Abstract:Objectives] Telomerase activity was studied in condyloma accuminata(CA) of cervix to assess whether it was correlated with the HPV type. 60 patients were assayed for telomerase activity using telomeric repeat amplification polymerase chain reaction (TRAP)-ELISA. Human Papillomavirus Type 6,11,16 and 18 Fluorescence Polymerase Chain Reaction(PCR) Diagnostic kit was used to test HPV type in CA. Raised telomerase activity was detected in condyloma acuminata. 24(40%) were HPV16/18 positive, 34(56.7%) had HPV6/11 infection. HPV16/18 infection correlate with more active telomerase than the HPV6/11 in the lesions. [Conclusion] This suggests that telomerase is activated in condyloma acuminata of cervix and may be interacted with HPV, thus they represent potential novel therapeutic targets in condyloma acuminata.
