高晶体-高胶体渗透压混合液对缺氧/氧再灌损伤的内皮细胞容积和存活率的影响

摘    要:目的 :观察高晶体 高胶体渗透压混合液(HHS)对缺氧 氧再灌损伤的人脐静脉内皮细胞(HUVEC)容积和存活率的影响。方法 :取HUVEC株ECV 30 4 ,分为对照组和实验组。两组细胞先置于缺氧 (95 %氮气 ,5 %CO2 )环境中 8h ,然后将实验组细胞置于HHS 15min ,再在常氧状态下 (95 %空气 ,5 %CO2 )培养 16h。对照组细胞直接置于常氧状态下 (95 %空气 ,5 %CO2 )培养 16h。两组细胞分别于实验前、实验开始后 15、6 0min和 8、8.2 5、16及2 4h测其细胞容积和存活率。结果 :对照组细胞在缺氧 6 0min后及氧再灌过程中明显肿胀 (P <0 .0 5或 0 .0 1) ,细胞存活率进行性降低 (P <0 .0 5或 0 .0 1)。实验组细胞经HHS处理后细胞容积较同一时段的对照组细胞容积明显缩小 (P <0 .0 1) ,存活率尽管仍低于实验前 (P <0 .0 1) ,但明显高于同一时段的对照组 (P <0 .0 5 )。结论 :HHS具有刺激内皮细胞产生延迟性调节性容积减少 ,保持容积稳定的作用 ,能在一定程度上降低内皮细胞的死亡率 ,保护缺氧 氧再灌损伤的内皮细胞。

Effect of hypertonic-hyperoncotic solution on cell volume and survival rate in human umbilical vein endothelial cells suffering from hypoxia/reperfusion
Abstract:AIM: To investigate the effect of hypertonic-hyperoncotic solution (HHS) on cell volume and survival rate in human umbilical vein endothelial cells (HUVEC) suffering from hypoxia/reperfusion. METHODS: HUVEC of ECV-304 were selected and randomly divided into control group and experimental group. Cells in two groups were cultured in hypoxia environment (95% nitrogen, and 5% carbon dioxide) for 8 h, then in normal environment (95% air, and 5% carbon dioxide) for 16 h in control group, and in HHS for 15 min in experimental group. The cell volume and the survival rate were detected before experiment, 15 min, 60 min, 8, 8.25 , 16, and 24 h after experiment in two groups. RESULTS: In control group, cells became significantly inflated and the survival rate decreased in hypoxia period after lasting 60 min and hypoxia/reperfusion period (P< 0.05 or P< 0.01 ). In experimental group, cells became deflated significantly compared with control group at the same period (P< 0.01 ). The survival rate decreased, but significantly higher than that in control group at the same period (P< 0.05 ). CONCLUSION: HHS can delay RVD,keep cell volume steady, and decrease cell mortality suffering from hypoxia/reperfusion.
