益肾活血汤抗单侧输尿管梗阻大鼠肾间质纤维化作用及机制的研究

摘    要:目的 探讨益肾活血汤对大鼠肾间质纤维化的拮抗作用及机制.方法 将60只大鼠随机分为假手术组、模型组、益肾活血汤治疗组、贝那普利治疗组.制备单侧输尿管梗阻(UUO)大鼠模型,在术后第21日处死大鼠留取梗阻侧肾组织观察肾间质病理变化,并用免疫组化法测定肾组织中纤溶酶原激活剂抑制物-1(PAI-1)、转化生长因子-β(TGF-β)及结缔组织生长因子(CTGF)的表达,处死前留取24 h尿测定尿蛋白,心腔穿刺取血测定血清尿素氮(BUN)、血清肌酐(Scr).结果 益肾活血汤组24h尿蛋白、BUN、Scr水平、肾间质纤维化程度及PAI-1、TGF-β、CTGF的表达量均明显低于模型组(P<0.05),但与贝那普利组差异无统计学意义(P>0.05).结论 益肾活血汤对肾间质纤维化具有拮抗作用,其机制可能为通过减少PAI-1、TGF-β、CTGF等致纤维化因子在肾组织中的过度表达来抑制肾间质纤维化.

The Antagonizing Effect and Mechanism of Yishenhuoxue Decoction on Renal Interstitial Fibrosis of Unilateral Ureteral Obstruction Rats

Abstract:Objective:To investigate the antagonizing effect and mechanism of Yishenhuoxue decoction on renal interstitial fibrosis of unilateral ureteral obstruction (UUO) rats.Methods:Sixty SD rats were randomly assigned to shame operation group,operation model group,Yishenhuoxue decoction group and Benazepril group after unilateral ureteral obstruction.Rats were killed on the 21 day after operation.Renal tissues were examined by HE stain.Immunohistochemistry was performed for plasminogen activator inhibitor-1 (PAI-1),transforming growth factor-β (TGF-β) and connective tissue growth factor (CTGF) in renal interstitium.BUN,Scr and 24 hours urinary protein quantity were measured.Results:The fibrosis index values,the expression of PAI-1,TGF-β,CTGF protein and the level of BUN,Scr,24 hours urinary protein quantity in Yishenhuoxue decoction group were significantly lower than those in model group (P< 0.05).There were no significant differeces between Yeshenhuoxue decoction group and Benazepril group (P> 0.05).Conclusion:Yeshenhuoxue decoction possesses antagonizing effects on renal interstitial fibrosis by depressing the expression of PAI-1,TGF-β,CTGF protein.
