摘    要:目的从分子水平研究Rb基因对骨肉瘤生长的影响,为骨肉瘤的基因治疗提供依据.方法用脂质体将人工合成的Rb反义寡脱氧核苷酸转入有Rb表达的人成骨肉瘤细胞.免疫组化方法检测肿瘤中Rb蛋白表达,MTT法检测细胞转染前后的增殖情况.同时流式细胞术检测细胞周期.结果转染Rb反义寡核苷酸后肿瘤细胞的Rb蛋白表达明显下降,G1期细胞比例减少,细胞增殖增加.结论转染Rb反义寡核苷酸的细胞,Rb蛋白表达明显降低,并引起肿瘤细胞增殖增加.

Abstract:Objective] To study the effects of Rb gene on the growth of osteosarcoma cells. Methods] Rb antisense oligodeoxynucleotides(AS-ODN) was synthesized and transfected into MG-63 cell lines by lioposome amine. The expression of Rb protein was determined by immunohistochemical methods, and proliferation of the cells and cell cycle distribution was studied by MTT methods and flowcytometry respectively. Results] The expression of Rb protein in the transfected cells was decreased greatly. Cell proliferation was increased and G1 phase cells was reduced. Conclusion] Rb antisense oligodeoxyeucleotide with liposome may inhibit the expression of Rb and enhance the growth of osteosarcoma cells.
