摘    要:目的:观察二甲双胍对胰岛素抵抗大鼠IL-6表达的影响,探讨二甲双胍改善胰岛素抵抗的作用机制。方法:29只正常雄性大鼠随机分为:正常饮食对照组(n=9),高脂高糖喂养造模组(n=20)。喂养6周后,将造模成功的17只IR大鼠再随机分为两组:①模型组:继续予高脂高糖饮食;②二甲双胍组:高脂高糖喂养加二甲双胍300 mg/(kg.d)];6周干预后,观测胰岛素抵抗大鼠IL-6表达的变化。结果:高脂高糖饲养6周后,造模组FBG、FINS均高于对照组,ISI低于对照组(P<0.01)。与模型组比较,药物组血清IL-6、脂肪和肝脏组织IL-6 mRNA的表达均降低(P<0.05),ISI均升高(P<0.05)。结论:下调IL-6的表达可能是二甲双胍改善大鼠胰岛素抵抗的机制之一。

Abstract:Objective To investigate the effect of metformin on the expression of IL-6 in rats with insulin resistance,to explore the mechanisms that why metformin could improve the insulin resistance.Methods Twenty-nine male Sprague-Dawley rats were randomly assigned to two groups:The normal diet group(control group,n=9)and the high-fat/high-sucrose diet group(n=20).After six weeks,The seventeen rats with insulin resistance were randomly assigned to two groups:①continue the high-fat/high-sucrose diet(model group,n=9);②a high-fat/high-sucrose diet combined with metformin(metformin group,n=7).After six weeks of intervention,investigate the effect of metformin on the expression of IL-6 in rats with insulin resistance.Results After fed with the high-fat/high-sucrose for six weeks,compared with the control group,the serum level of FBG and FINS were significantly increased,and ISI was significantly decreased in the model group(P0.05).After six weeks of intervention,Compared with model group,the serum content of IL-6 and the expression of IL-6 mRNA in adipose and liver tissue were significantly lower(P0.05);the ISI was significantly higher in the metformin group(P0.05).Conclusion Metformin could lower the level of IL-6 which might be the mechanism that metformin could improve insulin resistance.
