中药对溴隐亭致流产模型血清PRL的影响

目的 :探讨 PRL在流产过程中的影响及中药的作用机理。方法 :按 Gafveis法改良溴隐亭致 SD大鼠流产模型 ,于孕 d6～ d8皮下注射溴隐亭 0 .1 2 5 mg/d。随机分为两组 :A组 (中药组 ) ,B组 (模型组 ) ,分别于孕 d1～ d1 2灌服中药及水 ,另设 C组 (正常妊娠组 ) ,D组 (酒精溶剂组 ) ,E组 (非妊娠组 )。A～ D组均于孕 d1 2处死 ,比较胚胎数 ,以放免法测各组不同时期血清 PRL浓度。结果 :A组流产率低于 B组 ( 2 2 .2 2 % vs70 .83 % ,P<0 .0 5 ) ,妊娠成功率高于 B组 ;A组胎仔数显著多于 B组( 1 1 4vs2 5 ,P<0 .0 1 )。各孕日龄血清 PRL水平较动情期明显升高 ( P<0 .0 5 ,0 .0 1 ) ,A组 PRL在妊娠后呈上升趋势 ,与 C组、D组比较不明显 ,B组呈持平或下降趋势。结论 :该模型具有低 PRL致流产的特征 ,补肾益气中药可通过妊娠生理性高 PRL维持妊娠

Effect of Chinese Herbs on Serum PRL in the Rat Model of Bromocriptine Induced Abortion

Objective:To study the role of PRL in abortion,and the mechanism of Chinese herbs in preventing abortion. Methods: Gafvels a borting model SD rats was given subcutaneous injection of 0.125 mg of bromocrip tine qd during days 6~8 of pregnancy. Then these rats were randomly divided into 2 groups. Group A was treated with Chinese herbs ig for 12 days since vaginal plug was observed. Group B was fed with distilled water during days 1~12 of pre gnancy. Group C was normal pregnant rats without treatment;Group D was pregnan t rats recived SC of 31 μL of 75% alcohol during days 6~8 of pregnancy; Group E was non-pregnant rats. Serum prolactin(PRL) level at day 4,7,10 and 12 of pr egnancy and during estrus in non-pregnant rats was measured by radio-immuno a ssay. Results:Abortion rats in group A,B,C,D were 22.2 2%,70.83% (group A vs group B, P<0.05), 8% and 4%, respectivly. Successful p regnant rats in group A,B,C,D were 77.78%,29.13% (group A vs B, P<0.05) 92% ,96% respectively. The number of embryo in group A,B,C,D was 234, 64 (group A vs B, P<0.01), 242 and 262 respectivly. Levels of serial serum PRL of group A, B ,C,D of prgnancy during days 4,7,10,12 were eleveted higher than that non-pre gnancy rats(group E) at estrous cycle (P<0.01). Levels of serum PRL of group s A,C,D during days 7,10,12 were elevated higher than that of group B during sam e days (P< 0.05). Conclusion: Low serum PRL leve l during gestation is related to embryo miscarriage. Chinese herbs can reverse t he aborting effect of bromocriptine by increasing serum PRL.
