白细胞介素-17对子宫内膜癌细胞株HEC-1-B基质金属蛋白酶-2,-9基因表达及肿瘤细胞侵袭转移的影响

摘    要:目的 探讨白细胞介素(interleukin,IL)-17对子宫内膜癌细胞株HEC-1-B基质金属蛋白酶(matric metalloproteinases,MMP)-2,-9基因表达及肿瘤细胞侵袭转移能力的影响.方法 选择对数生长期人子宫内膜癌细胞株HEC-1-B,在无血清培养基中加入白细胞介素-17终浓度分别为1 ng/mL,10 ng/mL,100 ng/mL和无血清培养(空白对照)进行干扰处理后,分别纳入干扰组(干扰组1、干扰组2、干扰组3)及对照组.采用免疫荧光细胞化学方法检测子宫内膜癌细胞株HEC-1-B是否表达白细胞介素-17特异性受体C (IL-17RC)蛋白.采用逆转录荧光定量PCR法检测不同浓度白细胞介素-17对子宫内膜癌细胞株HEC-1-B基质金属蛋白酶-2,-9 mRNA基因表达水平的影响.采用transwell小室检测不同浓度白细胞介素-17干预后,子宫内膜癌细胞株HEC-1-B侵袭转移能力的变化.结果 人子宫内膜癌细胞株HEC-1-B高表达特异性白细胞介素-17受体C.白细胞介素-17各浓度干扰组(干扰组1,干扰组2,干扰组3)与对照组比较,干扰组基质金属蛋白酶-9 mRNA表达增加,差异有显著意义(P<0.01),且各浓度干扰组表现呈浓度依赖;而干扰组基质金属蛋白酶-2 mRNA表达无明显变化;干扰组穿膜细胞数明显增多,与对照组比较,差异有显著意义(P<0.01),且各浓度干扰组表现呈浓度依赖;而干扰组2与干扰组3穿膜细胞数较干扰组1增多,差异有显著意义(P<0.05).结论 白细胞介素-17能增强基质金属蛋白酶-9基因表达,促进子宫内膜癌细胞侵袭转移.
