脑胶质瘤动物的模型建立及cyclin D1表达的观察

摘    要:目的:建立类似人脑胶质瘤动物模型,观察cyclin D1在肿瘤组织中的表达.方法:借助动物立体定向仪,将体外培养大鼠C6胶质瘤细胞接种于Wistar大鼠左侧尾状核区.接种后观察实验大鼠生存状态,分别于接种后7,15d进行MRI检查.对每例荷瘤鼠死后进行解剖,行组织病理学和GFAP免疫组化检查.随后对不同存活期大鼠cyclin D1表达进行检测.结果:该方法建立的胶质瘤动物模型在组织病理学上接近人脑胶质瘤.而且具有颅内生长稳定,成瘤率高,重复性好.在每例胶质瘤组织中cyclin D1均呈阳性表达,主要在细胞胞核表达,不同存活期荷瘤鼠其瘤组织中cyclin D1表达量不同,存活时间较长者,其表达量明显减少.结论:此类模型肿瘤生长特性及病理特征与人脑胶质瘤相似.cychn D1表达量与脑胶质瘤大鼠存活时间呈负相关。

Establishment of a rat C6 brain-glioma model and observation of its cyclin D1 expression
Abstract:AIM: To explore the expression of cyclin D1 in the tumor tissues of glioma models. METHODS: Wistar C6 brain glioma models were established with stereotactic technique. The C6 cells cultured in vitro were stereotaxically implanted into the left caudate nucleus of Wistar. After C6 inoculation, an MRI protocol was used to evaluate the growth of the implanted C6 rat gliomas on day 7th and 15th. The living and survival time of animals with glioma was also observed. Tumor was confirmed with HE and GFAP immunohistochemistry staining and cyclin D1 was detected by immunohistochemistry technique. RESULTS: Rat C6 gliomas resembled human glioblastoma in the histopathological features. The expression of cyclin D1 was positive in glioma tissues. Immunohistochemical reaction of cyclin D1 was confined to the nuclei. A little difference of cyclin D1 expression was found in different survival time of the rats with tumors .The longer the rats survived, the less cyclin D1 was expressed. CONCLUSION: Our data suggest that the amplification and increased expression of cyclin D1 are negatively correlated with the survival time of the rats with tumors. The C6 glioma model of rats resembles human glioblastom in the histopathological features and can be used in studying the etiology of glioma.
