别嘌呤醇对缺血再灌注心肌MMP-1的变化的影响

摘    要:目的:研究缺血再灌注时的心肌MMP-1的表达变化及别嘌呤醇对其影响。方法:应用中国大白兔建立MI/R模型。设立:MI/R组、别嘌呤醇组、假手术组。用免疫组化法检测心肌中MMP-1的表达。用Maclab生理记录仪记录心功能变化。TBA法检测血液及心肌中MDA水平变化。结果:Sham组心肌MMP-1的表达极少。在缺血再灌注心肌中有MMP-1的表达,别嘌呤醇组较MI/R组明显减弱。心功能结果示再灌注4 h后MI/R组及别嘌呤醇组LVDP及±LVdp/dtmax均较SHAM组明显降低。但别嘌呤醇组LVDP及±dp/dtmax水平较I/R组高(P〈0.05)。MI/R组血液及心肌MDA水平高于别嘌呤醇组(P〈0.05)。结论:I/R后心肌中MMP-1的表达可能参与了再灌注损伤的过程。别嘌呤醇能抑制I/R时心肌MMP-1的表达并对心肌I/R损伤有保护作用。

The Changes of MMP-1 During Myocardial Ischemia/reperfusion and the Effect of Allopurinol on it

Abstract:Objective :To investigate the changes and significance of MMP - 1 in myocardium of rabbits during myocardial ischemia - reperfusion (MI/R) and the Protective effects of allopurinol on ischemia/reperfusion injury in rabbits hearts. Methods:Domestic rabbits were anesthetized and subjected to 45 rain of myoccardial ischemia followed by 4 h - reperfusion. Rabbits were randomly treated with allopurinol 2 days before operation. The myocardial function were assessed by The left ventricle development pressure(LVDP) , the highest rate of rise/decrease of left ventricular pressure ( ± dp/dtmax). The exprssion of Matrix metalloproteinase - 1 ( MMP - 1 ) protein in myocytes were observed using immunohistochemistry, and the density of its products was determined by image analysis. Myocardial malondialdehyde(MDA) content and the change of blood levels of MDA was observed with Biochemical techniques. Results:(1)LVDP, + dp/dtmax were significantly better in Allopurinol group than that in the I/R group( P 〈 0. 05 ). (2)Administration of allopurinol reduced the expression of MMP - 1 during I/R. (3) The content of MDA in serum and in myocardium was increased in I/R than Sham, and decreased in drug intervention groups than I/R group after reperfusion ( 2. 11 ± 0. 27 vs 1.84 ± 0. 27 nmol/mL, P 〈0. 05)Conclusions:MMP- 1 gene expression is upregulated in the I/R myocardium of rabbits arjd may be a important Inflammatory cytokine. As a effective composition, allopurinol may reduce the ischemia - reperfsion injury.
