臂丛根性撕脱伤后脊髓前角神经干细胞移植分化

摘    要:目的 观察臂丛根性撕脱伤后在脊髓前角移植神经干细胞的存活、迁移、分化情况。方法 取新生鼠脊髓。分离获得脊髓源性神经干细胞,在体外培养、扩增、鉴定,并用5-溴2-脱氧尿苷(Brdu)标记。取SD大鼠40只。随机分成臂丛神经根性撕脱伤移植灭活神经干细胞组(对照组)、臂丛神经根性撕脱伤移植神经干细胞组(实验组)。先将C5~C7神经根经后路撕脱,实验组把体外培养的神经干细胞移植于损伤侧C6脊髓前角,对照组则移植灭活神经干细胞。术后1,2,4,8,12周取脊髓标本进行组织学与免疫组织化学染色观察。结果 臂丛根性撕脱伤后在脊髓前角移植神经干细胞不仅能存活,并可向二端迁移达一个脊髓节段,分化为神经元以及星型胶质细胞。结论 神经干细胞在植入臂丛根性撕脱伤的脊髓节段前角后不仅能存活。并能迁移和分化。

Abstract:Objective To study the survival, migration and differentiation of the transplanted neural stem cells after brachial plexus avulsion in ventral horn of spinal cord. Methods Neural stem cells isolated from spinal cord of neonatal rats were cultured, expanded, labeled by BrdU before transplanted. Fourty adult healthy SD rats were randomly divided into experimental group (C_ 5-7 brachial plexus avulsion) and healthy control group, preformed as the model of brachial plexus avulsion(Roots C_ 5-7). Alive or dead stem cell were thansplanted into the C_6 ventral horn of spinal cord and histology and immunohistochemistry assay were carried out in the spinal cord at 1, 2, 4, 8, 12 weeks postoperatively. Results Transplanted neural stem cells can survive, migrate about one segment of spinal cord and differentiate into neurons and astrocytes. Conclusion Neural stem cells can survive, migrate and differentiate after transplanted into ventral horn of spinal cord in the rats which suffered from brachial plexus avulsion.
