原发性肝细胞肝癌中核不均一核糖核蛋白A1的定位及临床意义

摘    要:目的:观察核不均一核糖核蛋白A_1(heterogeneous nuclear ribonucleoprotein A_1,hnRNP A_1)在原发性肝细胞肝癌(肝癌)中的定位,初步分析该蛋白定位与肝癌患者临床病理特征之间的相关性及其预测患者术后复发转移的临床价值。方法:由323例肝癌患者肝癌组织构成组织芯片集,采用免疫组织化学染色技术,观察肝组织中hnRNP A_1的定位,分析hnRNP A_1定位与临床病理特征及患者预后的相关性。结果:hnRNP A_1蛋白在正常肝细胞核中表达呈弱阳性,在肝癌组织的细胞核和细胞质中均有不同程度的阳性表达。hnRNP A_1在肝癌细胞质中定位的肝癌患者的术后预后最差。结论:hnRNP A_1蛋白是较好的独立预后指标,其能准确预测肝癌患者术后的预后。

Abstract:Abstract: Background & aims: Heterogeneous ribonucleoprotein A1 (hnRNP A1) is a multifunctional factor which is dysregulated in tumorigenesis and tumor progression. The main objective of this study is to investigate the location of hnRNPA1 and its prognostic significance in hepatocellular carcinoma (HCC). Methods: The location of hnRNPA1 was studied in an independent cohort of 323 HCC patients by immunohistochemistry. Results: For hnRNPA1, there was weak-to-moderate intensity immunoreactivity with nuclear expression in normal liver, while showed cytoplasmic and nuclear loaction in HCC. Statistical analysis revealed that exclusively cytoplasmic hnRNPA1 accumulation is significantly correlated with poor prognosis of HCC patients. Conclusions: It is possible that the localization of hnRNPA1 may participate in the regulation of tumor progression.
