肝郁气滞证乳腺增生大鼠血中激素水平的变化

目的:建立中医肝郁气滞证乳腺增生大鼠模型,同时观察其激素水平的变化。方法:利用慢性多种不可预知的应激刺激随机组合,同时给予雌性大鼠肌肉注射苯甲雌二醇和黄体酮造模。通过放射免疫法测定各组大鼠静脉血中的雌二醇、孕酮、泌乳素、睾酮和促肾上腺皮质激素含量。结果:单纯注射苯甲雌二醇和黄体酮-乳腺增生组、单纯给予应激刺激-肝郁气滞组,既注射苯甲雌二醇和黄体酮又给予应激刺激-肝郁气滞证乳腺增生组与正常对照组比较,血清中E2、PRL、ACTH水平明显升高;P、T含量明显降低。结论:利用慢性多种不可预知的应激随机组合,同时注射苯甲雌二醇和黄体酮造模,通过观测血中激素水平的变化,可较好地模拟中医肝郁气滞证乳腺增生病激素水平改变的表现,初步建立肝郁气滞证乳腺增生大鼠模型。

The Hormone Level Change of the Hyperplastic Mammary Glands Rats by Stagnant Qi Due to Depression of the Liver

Objective：To establish hyperplastic mammary glands rats by stagnant Qi and observe the change of hormone level due to depression of the liver. Methods： To apply to the combination of chronic various of unpredictable stress stimulation stochastic, simultaneously inject intramuscular estradiol benzoate and the progestin to the female rats. Lastly, estradio 1（E2）, the progesterone（P）, prolactin （PRL）, the testosterone （T） and Adrenocorficotrophic （ACTH） were tested by radioimmunoassay （RIA）.Results： The testers inject estradiol benzoate and the progestin merely-the hyperplastic mammary glands rats group （EP）; secondly, we only give the stress stimulate-stagnant Qi due to depression of the liver rats group （S）; thirdly, we not only inject estradiol benzoate and the progestin to the rats but also give the stress stimulation-the hyperplastic mammary glands rats by stagnant Qi due to depression of the liver group （EP＋S）.All groups are compared with the normal control group： the serum E2, PRL, ACTH level elevates obviously; P,T content reduces obviously. Conclusion： The performance of Chinese medicine of hyperplastic mammary glands by stagnant Qi due to depression of the liver might be imitated better after employing the combination of chronic various of unpredictable stress stimulation stochastic and injecting intramuscular estradiol benzoate and the progestin to the female rats. The model will be established initially.
