多西他赛联合顺铂治疗蒽环类化疗失败乳腺癌的临床研究

目的 研究多西他赛联合顺铂治疗蒽环类化疗失败的复发转移性乳腺癌的疗效及不良反应。方法 40例蒽环类化疗失败的复发转移性乳腺癌患者，采用多西他赛联合顺铂治疗，多西他赛37．5mg／m^2静滴第1、8天；顺铂75．0mg／m^2静滴第1-3天分次给予。每3周为1周期，每周期评价疗效并记录不良反应。结果 40例患者均可评价疗效，完全缓解（CR）4例，部分缓解（PR）17例，有效率（CR＋PR）52．5％。全组共化疗152周期，无化疗相关死亡患者，主要不良反应为消化道反应和骨髓抑制，Ⅲ-Ⅳ度呕吐12．5％，Ⅲ-Ⅳ度腹泻5．0％，Ⅲ—Ⅳ度中性粒细胞减少17．5％。结论 多西他赛联合顺铂是治疗蒽环类化疗失败的复发转移性乳腺癌的有效方案，不良反应可以耐受。

Clinical study of Docetaxel combined Cisplatin in treatment of Anthracycline-pretreated metastatic breast cancer

[Objective] To evaluate the efficacy and toxicity of Docetaxel-Cisplatin combination chemotherapy on Anthracycline-pretreated metastatic breast cancer.[Methods] Forty patients with metastatic breast cancer failed to prior treatment of Anthracycline,were treated with Docetaxel and Cisplatin combination regimen.Docetaxel 37.5 mg/m2 ivgtt,d1,8,Cisplatin 75 mg/m2 ivgtt in 3 days,once every 3 weeks.Efficiency and toxicity were observed in every cycle.[Result] Forty patients were evaluated for response.The overall response rate was 52.5%,with 4 CR,17 PR.A total of 152 cycles were administered.There was no death associated with chemotherapy.Major toxicities were gastrointestinal toxicity and myelosuppression with grade III-IV vomiting in 12.5%,grade III-IV diarrhea in 5.0%,grade III-IV neutropenia in 17.5%.[Conclusion] The combination of Docetaxel and Cisplatin is an effective regimen for patients with metastatic breast cancer who failed Anthracycline-pretreatment.The toxicities are tolerable to the majority of patients.
