可逆性上矢状窦血栓大鼠模型的建立

摘    要:目的建立可逆性上矢状窦血栓(SSST)大鼠模型,观察模型的稳定性和成功率。方法取雄性SD大鼠90只,随机分为模型组和对照组,每组45只;模型组采用上矢状窦局部置管并注射活化部分凝血活酶试剂建立大鼠SSST模型;对照组采用上矢状窦三氯化铁滤纸贴敷法建立SSST模型。两组于手术后24h行MRI及MR静脉造影(MRV),确定造模是否成功。模型成功后,分别于1、2、3、4周行MRI、MRV,观察上矢状窦再通情况及脑组织改变;取大鼠上矢状窦顶叶皮质脑组织标本行病理学观察。结果模型组模型成功41只,成功率为91%;总再通率为10%;对照组模型成功34只,成功率为75%,总再通率为45%。两组比较差异有统计学意义,P〈0.05。模型组大鼠脑组织水肿及梗死较对照组明显,而内皮细胞损伤较对照组轻微。结论改良法建立的大鼠可逆性上矢状窦血栓模型成功率高、稳定和重复性好。

Abstract:Objective To modify the rat model of reversible superior sagittal sinus thrombosis(SSST) and to observe its stability and successful rate.Methods Ninety male Sprague-Dawley rats were randomly allocated into a modified group,and a control group(n=45 in each group).The model of SSST in modified group was established by using superior sagittal sinus local intubation and injecting activated partial thromboplastin regent and the model of SSST in the control group was induced by topical application of superior sagittal sinus ferric chloride filter paper.MRI and magnetic resonance venography(MRV) were performed in both groups within 24 hours after the operation,and the successful rate of the models was determined.MRI and MRV were performed at 1,2,3 and 4 weeks respectively after the models were established successfully,and the recanalization of superior sagittal sinus and the changes of brain tissues were observed.The rat SSST and brain tissue samples were obtain for pathological observation.Results The successful rate in modified group was 91%(41 rats),and the total recanalization rate was 10%;the successful rate in control group was 75%(34 rats),and the total recanalization rate was 45%.There were significant differences between the two groups(P<0.05).Brain edema and infarction in the modified group was more obvious than those in the control group,but endotheliocyte injury in the modified group was milder than that in the control group.Conclusion The rat model of reversible SSST established by the modified method has higher successful rate,and the model stability and reproducibility are good.
