合成鱼腥草素对巨噬细胞呼吸爆发、细胞内游离钙离子浓度及T细胞分泌白细胞介素-2的影响

摘    要:目的为了进一步分析合成鱼腥草素的免疫调节作用机理 ,研究合成鱼腥草素对于巨噬细胞呼吸爆发、细胞内钙离子浓度以及T细胞分泌白细胞介素水平的影响。方法巨噬细胞分离自大鼠腹腔灌洗液。以 2′ ,7′ 二氯荧光素二乙酯作为荧光指示剂 ,采用流式细胞术检测巨噬细胞的呼吸爆发。以fura 2作为钙离子荧光指示剂采用荧光分光光度法测定细胞内钙离子浓度。利用淋巴细胞分离液采用密度梯度离心法分离外周血T细胞 ,并用尼龙毛柱加以纯化。用ELISA法测定在亚适剂量ConA、白细胞介素 1α(IL 1α)以及白细胞介素 1β(IL 1β)存在的条件下合成鱼腥草素对白细胞介素 2 (IL 2 )分泌的影响。结果合成鱼腥草素可以刺激巨噬细胞呼吸爆发 ,提高细胞内钙离子浓度水平 ,促进T细胞分泌IL 2。结论合成鱼腥草素可能具有激活巨噬细胞和T淋巴细胞的作用 ,从而部分解释合成鱼腥草素的佐剂作用以及治疗感染性疾病的作用机理。

Abstract:Objective To investigate the effects of sodium houttuyfonate on respiratory burst and intracellular free calcium concentration in macrophages and the secretion of IL 2 from T cells.Method Macrophages were collected and separated from the fluid of the peritoneal cavities of animals.Respiratory burst of macrophages was investigated by flow cytometry.Intracellular free calcium concentration was tested by spectrofluorophotometry using fura 2 as the calcium probe.T cells separated from the peripheral blood of animals by density gradient centrifugation using lymphocyte separation medium and enriched by nylon wool column chromatography were incubated with or without sodium houttuyfonate in the presence of concanavalin A,IL 1 α and IL 1 β ,and the secretion of IL 2 was assayed by the ELISA method.Result It was observed in the study that sodium houttuyfonate treatment induced respiratory burst and increased the concentration of free calcium in macrophages.It was also observed that the secretion of IL 2 from T cells was greatly increased by the sodium houttuyfonate treatment.Conclusion These findings indicate that sodium houttuyfonate may have the activity to activate macrophages and T cells.
