摘    要:目的:探讨缩泉润肠方对便秘模型大鼠结肠黏膜水通道蛋白的影响.方法:采用复方地芬诺酯混悬液(100 mg·kg-1)连续灌胃3周建立大鼠便秘模型.将造模成功的大鼠随机分模型组、乳果糖口服液组(0.670 g·kg-1)、缩泉润肠颗粒剂低剂量组(14.04 g·kg-1)、缩泉润肠颗粒剂高剂量组(28.08 g·kg-1)...

Abstract:Objective: To explore the effect of Suo Quan Run Chang Formula on aquaporin in the colonic mucosa of aged rats with constipation.Methods: The constipation rat model was established by intragastric administration of(100 mg·kg-1) compound diphenoxylate suspension for 3 weeks.The successfully modeled rats were randomly divided into the model group, lactulose oral liquid group(0.670 g·kg-1),low-dose Suo Quan Run Chang Granules group(14.04 g·kg-1),and high-dose Suoquan Runchang granules group(28.08 g·kg-1),10 rats in each group, and another 10 blank control rats were given the corresponding drugs for 2 weeks.The carbon ink advancing rate and fecal water content of rats in each group were observed.RT-PCR method was used to detect the expression of AQP3 mRNA,AQP8 mRNA,and AQP9 mRNA in the colon mucosa of rats.HE staining was used to observe the changes of pathological damage in the colon of rats.Results: Compared with the model group, the carbon ink advancing rate and fecal water content of the low and high dose groups of Suo Quan Run Chang Granules increased significantly(P<0.05).In the lactulose oral liquid group, the carbon ink advancement rate was not significantly improved(P>0.05),and the fecal water content was significantly increased(P<0.05).The levels of AQP3 mRNA and AQP8 mRNA in the low-dose and high-dose groups of Suoquan Runchang Granules and the lactulose oral liquid group decreased significantly, and the levels of AQP9 mRNA were significant(P<0.05).Compared with the lactulose oral liquid group, the carbon ink advancement rate and fecal water content of the low-dose and high-dose groups of Suoquan Runchang Granules were significantly increased(P<0.05).The levels of AQP3 mRNA and AQP8 mRNA in the low-dose and high-dose groups of Suo Quan Run Chang Granules were significantly reduced, and the levels of AQP9 mRNA were significantly increased(P<0.05).And the colon tissue of the Suoquan Runchang Granules group was improved.Conclusion: Suo Quan Run Chang Formula may regulate the secretion and absorption of intestinal water, increase fecal water content, accelerate colonic peristalsis, and exert a laxative effect.
