白花蛇舌草诱导HSP70表达对H22肝癌细胞移植瘤的干预作用

摘    要:目的观察白花蛇舌草诱导HSP7。表达对H22肝癌细胞移植瘤小鼠病理改变及T细胞亚群的影响。方法将中药处理后的H22肝癌细胞免疫小鼠采用HSP70单抗封闭技术分为白花蛇舌草未封闭、封闭,党参未封闭、封闭及RPMI-1640空白对照共5组,观察各组移植瘤小鼠瘤组织、胸腺及脾脏的病理改变及外周血CD4^+、CD8^+表达情况。结果白花蛇舌草未封闭组肿瘤组织坏死较为明显,胸腺皮质及脾脏白髓增厚,可见大量巨噬细胞;白花蛇舌草封闭组,党参未封闭及封闭组肿瘤组织的坏死、胸腺及脾脏的病理改变与空白对照组比较除脾脏可见少量巨噬细胞外,未见其他明显不同。白花蛇舌草未封闭、封闭和党参未封闭、封闭组CD3^+的表达均较空白对照组增高;与空白对照组比较,白花蛇舌草未封闭组CD8^+、封闭组CD4^+表达明显上调。党参未封闭及封闭组CD4^+、CD8^+表达与空白对照组比较也有所增高,但差异无显著性意义。结论经白花蛇舌草处理的H22肝癌细胞能一定程度提高小鼠对同种移植瘤的主动免疫抗瘤作用,其机理可能与诱导肝癌细胞HSP7。表达,增强其免疫原性有关。

Abstract:Objective To observe the influence of HSP_(70)expression induced by Herba Hedyotidis Diffusae on the patho- logical changes and T lymphocyte subgroup in mice with H22 cells transplanted tumor.Methods Mice with peritoneal transplantation of H22 liver cancer ceils were treated with Herba Hedyotidis Diffusae(HHD)and Radix Codonopsis(RC) by gastric gavage respectively for 12 days.Then H22 cells isolated from the mice treated by herbal medicine were subcu- taneously injected into mice right axilla.The mice immunized by right subaxillary injection were divided into 5 groups: HHD group(immunized with HHD-treated H22 cells),blocked HHD group(immunized with HHD-treated H22 cells blocked by HSPTo monoclonal antibody),RC group(immunized with RC-treated H22 cells),blocked RC group (immunized with RC-treated H22 cells blocked by HSPT0 monoclonal antibody)and blank control group(immunized with RPMI-1640).The pathologic changes of tumor tissue,thymus gland and spleen,as well as the expression of peripheral blood CD4~+ and CD8~+ in the mice were observed.Results As compared to the blank control group,there existed obvi- ous necrosis of the tumor tissue,thickened thymic cortex and spleen white pulp and a large amount of macrophagocytes in HHD group,while there only existed a few macrophagocytes in blocked HHD group,RC group anti blocked RC group. Compared with the blank control group,CD3~+ expression in other four groups were increased ,CD4~+ and CD8~+ expres- sion in HHD group and blocked HHD group were up-regulated obviously,CD4~+ anti CD8~+ expression in RC group and blocked RC group were increased also,but the difference was insignificant.Conclusion H22 liver cancer cells pretreat- ed by HHD can increase mice active immunity on the same kind of transplanted tumor.The machnism may be related to the induction of the HSP_(70)expression in H22 liver cancer cells and the reinforcement of its immunogenicity.
