根霉脂肪酶催化合成单甘酯的研究

摘    要:采用聚氨酯类材料为载体固定化根霉BUCT-11发酵生产脂肪酶.对根霉BUCT-11游离脂肪酶催化反应的工艺条件进行了优化,采用双温程等手段以后,产物中单甘酯质量分数可达47.5%.固定化细胞简单处理后,用于甘油解反应,在最佳条件下双温程后,单甘酯质量分数达到35.5%.文中实现了根霉BUCT-11发酵产物的综合利用,降低了单甘酯合成中酶的成本,因而具有良好的工业应用前景.

Abstract:Lipase was produced by immobilized Rhizopus BUCT211 with polyurethane. Both the lipase and immobilized cell were used for synthesis of monoglyceride. Among several lipases f rom different sources the lipase of Rhizopus BUCT211 was found to be suitable for synthesis of monoglycerides. The optimization of glycerolysisconditions of palm oil by f ree Rhizopus BUCT211 lipase was made. Under the optimal conditions(enzymatic activity : 315 U/ g palm oil , temperature : 30 ℃, water content in glycerol : 4100 % and reaction time : 24 h) the MG yield achieved 37.14 % , and by a two2step temperature reduction the monoglyceride content increased to 47.15 %. After simply pret reated the immobilized cells were also used to catalyze the glycerolysis reaction of palm oil. By study of the glycerolysis activity of the immobilized cells , the optimal condition (temperature : 28 ℃, celladded : 016 g/ g palm oil , water content in glycerol : 3100 % and reaction time : 36 h) was attained under which the MG yield arrived at 2910 % , and by a two2step temperature reduction 3515 % monoglyceride was achieved.
