ERCC1表达水平与晚期非小细胞肺癌患者化疗疗效的相关性

摘    要:目的:研究Ⅲb、Ⅳ期非小细胞肺癌(NSCLC)患者肿瘤组织内核苷酸切除修复交叉互补组1(excision repair cross complementation group1,ERCC1)基因mRNA表达水平与含铂类方案化疗疗效的相关性。方法:符合入选条件的患者56例,均为Ⅲb、Ⅳ期患者。所有的患者经皮肺穿刺或经纤支镜活检病理证实为NSCLC。化疗前利用RNA提取技术及RealtimeRT-PCR技术对其组织学标本进行ERCC1 mRNA相对定量分析。患者分别给予4~6个周期的含铂类方案化疗(吉西他滨/顺铂、紫杉醇/顺铂方案),对患者化疗后的总生存期(overall survival,OS)、肿瘤进展时间(time to progression,TTP)及化疗后反应率(response rate,RR)进行评估。结果:56例患者活检标本的ERCC1 mRNA相对于内控管家基因GAPDH mRNA表达的平均表达水平为6.1×103。以ERCC1 mRNA表达水平≥6.1×103的22例患者作为高表达组,以ERCC1 mRNA表达水平<6.1×103的34例患者作为低表达组。高表达组患者化疗后RR、OS及TTP...
