摘 要: | 目的:探讨非小细胞肺癌(non—smallcelllungcancer,NSCLC)组织中乙酰辅酶A羧化酶磷酸化后的产物(ace—tyl—coacarboxylaseproductofphosphorylation,P-Acc)与核转录因子KB(nuclearfactorkappaB,NFKB)的表达和相关性及其与临床病理因素的关系。方法:以赣州市肿瘤医院(赣州市肿瘤防治研究所)2008-01—012010—123l有完整临床病理资料,病理检查确诊为NSCI。C的62例癌组织作为研究组,20例同期因其他原因行肺叶切除患者的正常肺组织作为对照组。免疫组化SP法检测P—ACC和N卜KB的表达情况。结果:P—ACC在正常肺组织中80.0%(16/20)阳性表达,在NSCLC组织中33.9%(21/62)阳性表达,差异有统计学意义,P=0.0001;NF-xB在正常肺组织中15.0%(3/20)阳性表达,在NSCI。C组织中75.8%(47/62)阳性表达,差异有统计学意义,P〈0.05。在NSCI。C组织中,P-ACC的阳性表达与肿瘤大小显著相关,P=0.033;而与淋巴结转移、临床分期及病理类型无关,P〉0.05。NF—xB的阳性表达与肿瘤大小、淋巴结转移及临床分期无关,P〉0.05;而与病理类型显著相关,P=0.008。P—ACC与NF_KB的阳性表达之间呈显著负相关,r=-0.409,P〈O.001。结论:P-ACC阳性表达降低可能激活NF-xB阳性表达,促进NSCLC的发生、发展及侵袭转移。

Abstract: | OBJECTIVE: To explore the expression of acetyl-coa carboxylase product of phosphorylation (P ACC) and nuclear factor kappaB(NF-kB) in non small cell lung cancer(NSCLC) tissue and the relationship of the tumor size, lymph node metastasis, clinical stage and pathological type, and study the correlation of them. METHODS: 62 cases of NSCLC tissue were as a patient group. Twenty cases normal lung tissues as controls. Immunohistochemical SP method was used in detection of the expression of P-ACC and NF-kB in lung cancer and normal lung tissue. The immunohisto chemical SP expression of P-ACC and NF-kB was investigated in 60 NSCLC lung cancer tissue and 20 lung normal tissue because of other reasons for lobectomy. RESULTS: PACC 80.0% (16/20) positive expression in normal lung tissue and 33.9%(21/62) positive expression in NSCLC tissues,the difference was significant (P=0. 000 1). NF-eB was 15% posi- tive expression in normal lung tissue and 75.8%(47/62) positive expression in NSCLC tissues,the difference was signifi- cant (P〈0. 05). In NSCLC, the positive expression of P-ACC was associated with tumor .size significantly (P = 0. 033), but it was no correlation between lymph node metastasis,clinical stage and pathological type. We also found that there was no correlation between the positive expression of NF-kB and tumor size, lymph node metastasis, clinical stage but there was significant correlation with pathological type (P=0. 008). Further analysis revealed that the positive expression of P- ACC and NF-eB in NSCLC was significantly negative correlation (r= - 0. 409, P〈0.001 ). CONCLUSION: The reduced positive expression of P-ACC may activate positive expression of NF-kB, and promote the occurrence, development, inva- sion and metastasis of NSCLC. |
