Abstract:Objective To investigate the effects of Huayu Xiaozheng compound on the apoptosis and invasion of trophoblast cells in tubal pregnancy and the regulation mechanism of PI3K/Akt/mTOR signaling pathway.Methods Different concentrations of Huayu xiaozheng compound water extract were used to interfere with tubal pregnancy trophoblast cells.The blank group,western medicine group(methotrexate),and the control group(phosphatidylinositol 3-kinase inhibitors LY294002 and mammalian taget of rapamycin inhibitors rapamycin,were set up.Flow cytometry was used to detect the apoptotic rate after 72 hours intervention.Transwell assay was used to detect the cell invasion ability.Western-blot was used to detect the protein expression level of PI3K/Akt/mTOR signaling pathway.Results The apoptosis rates were up-regulated in all groups.The apoptosis rate increased with the increase of Chinese medicine concentration,and the promotion of apoptosis was more obvious in the high-dose Chinese medicine group than in the western medicine group(P<0.05).The number of cell transmembrane could be down-regulated in each treatment group.With the increase of Chinese medicine concentration,the cell number of transwell invasion cabin decreased and the invasion ability was weakened.There was no significant difference in the effect of high dose Chinese medicine group and western medicine group on cell invasion ability(P>0.05).The expression levels of p-Akt and p-mTOR protein in cells of Chinese medicine groups decreased with the increase of concentration(P<0.05).Compared with inhibitors of upstream and downstream pathways,the difference was not statistically significant(P>0.05).Conclusion Huayu Xiaozheng compound promote the cell apoptosis and inhibit cell invasion of trophoblast in tubal gestational,and it is concentration dependent,which may be related to down-regulating the activity of PI3K/Akt/mTOR signaling pathway in cells.
