不同品种芒果叶水提物HPLC指纹图谱及聚类分析

摘    要:目的: 建立不同品种芒果叶水提物的HPLC指纹图谱,并进行聚类分析。与近缘品种扁桃叶指纹图谱进行分析比较,为其质量控制和药材鉴别提供依据。方法: 以默克STAR RP-18e (5 μm,4.6 mm×250 mm) 为色谱柱,流动相为乙腈-0.1%磷酸溶剂系统,梯度洗脱,检测波长216 nm,对各批样品色谱图采用聚类分析进行归类,并将相同条件下测得的扁桃叶图谱与之相比较,分析区别。结果: 确定了芒果叶水提物指纹图谱9个共有峰,运用聚类分析的方法将31个品种芒果叶分成三大类,相同条件下测得的扁桃叶指纹图谱与芒果叶指纹图谱成分及含量存在一定的差异。结论: 建立的芒果叶水提物HPLC指纹图谱及质量分析方法,精密度、重现性和稳定性较好,能较好地区分芒果叶与近缘品种扁桃叶,为有效控制芒果叶药材的质量提供了科学依据。

Abstract:OBJECTIVE To establish HPLC fingerprints of water extracts from mango leave of varied varieties, compare and establish the HPLC fingerprints of mango leaves and almond leaves for quality control. METHODS Merck STAR RP-18e (5μm,4.6×250 mm) column was used for separation,with acetonitrile-0.1% phosphoric acid as mobile phase for gradient elution. The detection wavelength was at 216 nm,the data were analyzed by the similarity of fingerprint software. The fingerprints would be compared with those obtained by using almond leaves under the same conditions. RESULTS Nine common peaks were identified in chromatograms of mango leaves, and mango leaves of 31 varieties were divided into 3 categories by using clustering analysis. Under the same conditions, fingerprints of almond and mango leaves showed varied ingredients and contents. CONCLUSION This method is accurate, reproducible and stable, which can be used to distinguish mango and almond leaves, and provide scientific basis for quality control of mango leaves.
