氨磷汀对豚鼠早期中耳放射损伤的保护与ICAM-1表达的相关性研究

摘    要:目的 通过检测氨磷汀与细胞间黏附分子1(ICAM-1)在放射性中耳炎中表达的相关性,探讨该病的发病机制。方法 38只豚鼠分成5组,A组2只,不加任何干预,B、C、D、E每组9只,印C07射线3GY/次,5次/周,总量45GY照射右耳,B组和D组为生理盐水组,C组和E组为氨磷汀组,C组照射完成后在第2天处死动物,D、E组在照射后第30天处死动物。以免疫组化法检测中耳粘膜ICAM-1,同时用扫描电镜观察中耳粘膜。结果 辐射对豚鼠中耳粘膜上皮有损伤作用,同时伴随ICAM-1的表达,照射前30分钟腹腔注射氨磷汀能使纤毛上皮受到保护。结论 ICAM-1的表达与中耳粘膜早期辐射损伤密切相关,氨磷汀对中耳粘膜早期辐射损伤有保护作用。辐射产生的自由基与中耳粘膜ICAM-1表达上调相关。咽鼓管狭窄可能是粘膜ICAM-1持续表达的原因之一。

Relationship Between the Effect of Amifostine and ICAM-1 Expression
Abstract:Purpose Aims of our study were to characterize the relationship between the effect of amifostine and the ICAM-1 expression in early phase radiation induced otitis media and to illustrate the possible machanism of early phase radiation induced otitis media. Methods Expression of ICAM-1 and scanning electron microscope examination of middle ear tissue were studied using 38 middle ear mucosa guinea pig modeles.Amifostine was administered intraperineally as an aqueous solution 30 min before every irradiation in groups C and E,sterile saline was administered intraperineally 30 min before every irradiation in another two groups B and D.No radiation was given in group A.Groups B and C were killed the next day.Groups D and E the 30 days after finishing the planned irradiation. Results ICAM-1 in middle ear mucosa was strongly expressed after administered 45GY dose,Enhanced ICAM-1 expression was accompanied by the pathomorphology of the middle ear tissue which was demonstrated by SEM.The ICAM-1 expression was stronger in the next day groups than in the 30 days groups after finishing the planed radiation dose.Amifostine provided protection from irradiation injury to the middle ear,which was accompanied by downregulation of the expression of ICAM-1 in the middle ear mucosa.There was still slight expression of ICAM-1 until 30 days later after irradiation. Conclusions lrradiation increased the expression of ICAM-1 in the middle ear mucosa.Amifostine provided protection from early irradiation injury to the middle ear.Oxygen free radicals derived from irradiation was related to the upregulation of ICAM-1 expression.Continuous ICAM 1 expression might be related to the stenosis of eustachian tube.
