黄芪多糖对Hhcy大鼠心肌缺血再灌注过氧化的影响

摘    要:目的:探讨黄芪多糖(Astragalus Polysacharin,APS)对高同型半胱氨酸血症(hyperhomecysteinemia,Hhcy)大鼠心肌缺血再灌注损伤的保护作用及机制。方法:应用腹腔注射2%L-半胱氨酸5 ml/kg复制Hhcy大鼠模型,10周后,腹腔注射戊巴比妥钠麻醉下气管插管安装呼吸机,开胸结扎冠状动脉左室降支90 min后松开结扎线再灌注90 min。标Ⅱ导联全程记录心电图,并于再灌注90 min取血。检测肌酸激酶(CK)、乳酸脱氢酶(LDH)、心肌肌钙蛋白Ⅰ(Tn-Ⅰ)、同型半胱氨酸(Hcy)、丙二醛(MDA)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)。结果:每天腹腔注射2%L-半胱氨酸5 ml/kg,10周后,血浆Hcy升高。黄芪多糖干预组与Hhcy模型对照组比较CK、LDH、Tn-Ⅰ水平明显降低,而CAT、SOD明显升高。结论:黄芪多糖对Hhcy大鼠心肌缺血再灌注损伤具有保护作用,其机制与抗过氧化密切相关。

Effect of Astragalus polysaccharide on the myocardial ischemia-reperfusion peroxidation of Hhcy rat

Abstract:Objective To explore injury protection function and mechanism of astragalus polysaccharide(Astragalus Polysacharin,APS) on myocardial ischemia-reperfusion of homocysteine(hyperhomecysteinemia,Hhcy) rat.Method We applied intraperitoneal injection of 2% L-cysteine 5 ml/kg to copy Hhcy rats models,10 weeks later,they had intraperitoneal injection of sodium pentobarbital anesthesia and trachea cannula,installed breathing machine,opened chest,ligated descending left ventricular of coronary artery after 90 min,loosened ligature and had reperfusion for 90 min.Mark Ⅱ guide league recorded the electrocardiogram(ecg),and took blood in reperfusion 90 min.Tested creatine kinase(CK),lactate dehydrogenase(LDH),myocardial troponin Ⅰ(Tn-Ⅰ),homocysteine(Hcy),malondialdehyde(MDA),catalase(CAT),superoxide dismutase(SOD).Results They had intraperitoneal injection of 2% L-cysteine 5 ml/kg,every day,after 10 weeks,plasma Hcy rose.CK,LDH,Tn-Ⅰ level of astragalus polysaccharides intervention group obviously reduced than Hhcy model,while CAT and SOD increased significantly.Conclusion Astragalus polysaccharide on myocardial ischemia-reperfusion injury of the Hhcy rat has protective effect,and the mechanism is closely related to the peroxide resistance.
