西安汉族人群HLA-DQA1基因座遗传多态性研究

目的　了解西安汉族群体HLA DQA1基因座的基因及基因型分布 ,获得西安汉族群体HLA DQA1群体遗传学数据。方法　采用PERKIN ELMER公司生产的AmpliTypePM试剂盒 ,PCR扩增HLA DQA1基因座 ,与 11个寡核苷酸探针杂交 ,对西安地区汉族 10 2名无关个体HLA DQA1基因座进行基因频率调查。结果　检出 7种等位基因、2 8种基因型 ,经 χ2 检验基因型的观察值与期望值符合Hardy Weinberg平衡定律 (P >0 .5 ) ,其杂合度 (H)、个体识别率 (DP)、非父排除率 (EPP)和多态信息量 (PIC)分别为 0 .8317、0 .9393、0 .786 6和0 .82 5 0。结论　所得到的群体遗传学数据为遗传学、法医学个体识别和亲权鉴定提供依据

A study on the polymorphism of HLA-DQA1 loci in Xi'an Han population

Objective To explore the HLA DQA1 allele and genotype frequencies of Xi'an Han population and obtain genetic data. Methods The results of HLA DQA1 typing of 102 randomly selected from Xi'an Han population were obtained by using the Ampli Type HLA DQ alpha forensic DNA amplification and typing kit. Results 7 alleles and 28 genotypes were detected in Xi'an Han population. HLA DQA1 loci matched Hardy Weinberg expectation. The heterogeneity was 0.8317, the discrimination power was 0.9393, the excluding probability of paternity was 0.7866, and the polymorphism information content was 0.8250. Conclusion The frequency data can be used in population genetic and forensic analyses and paternity tests to estimate the frequency of a DNA profile in the Chinese population.
