“卡宁”抗骨肿瘤的机理初探

摘    要:目的体外观察抗癌中药“卡宁”对人成骨肉瘤细胞株MG-63的凋亡诱导基因fas影响,探讨其抗肿瘤治疗的作用机制。方法采用MTT比色法观察含药血清对体外培养的人骨肿瘤细胞株的抑制能力,PCR检测对凋亡诱导基因fas的影响。结果体外实验显示中药组fas的活性明显高于对照组(p=0.038),MTT结果显示细胞生长抑制率与含药血清作用时间及浓度呈正相关,p<0.05,在15%含药血清72h时作用最强。体外实验表明,“卡宁”对MG-63细胞的fas活性有促进作用。结论对肿瘤细胞fas的促进可能是抗癌中药“卡宁”诱导肿瘤细胞凋亡作用机制的一部分。

Abstract:Objective To evaluate the effect of Chinese medicine “caning” by antiapotosis gene fas so as to detect the mechanism of treating cancer. Method We observed the inhibitive effect of drug-containing serum to human osteoma cell line by MTT colorimetry and its antiapoptosis effect of fas gene by PCR assay. Result It indicated that the fas activity of the Chinese herb group was dramatically higher than that of the control. It showed that the inhibition ratio of cell growth was positively related to the concentration and the time of the drug-containing serum, p<0.05. It worked the best in the 15% drug-containing serum. It showed that “canning” can promoted the fas activity of the MG-63 cells in vitro Conclusion Anticancer drug “canning” induced cell apoptosis by enhancing the fas gene.
