摘    要:目的 :研究咖啡因对低钾诱导的小脑颗粒神经元凋亡的保护作用与细胞内钙离子浓度升高之间的关系。方法 :小脑颗粒神经元培养 ,凝胶电泳和fura 2荧光技术测胞内钙离子浓度。结果 :低钾的促凋亡作用可被咖啡因浓度依赖性地保护 ,且咖啡因的保护作用不受ryanodine敏感性钙释放的阻断剂 (ryanodine、dantrolene)的影响 ;也不被L 型钙通道阻断剂 (硝苯地平、维拉帕米和尼莫地平 )和NMDA受体阻断剂 (MK80 1)抑制。结论 :咖啡因对低钾诱导的小脑颗粒神经元凋亡的保护作用并不依赖胞内钙离子浓度的升高。

Abstract:Objective: To study the relationship between the protective effect of caffeine and the level of intracellular calcium Method: Cerebellar granule neurons culture, agarose gel eletrophoresis and measurement of intracellular Ca 2+ using fura 2 Results: Culture cerebellar granule neurons died by apoptosis when switched from a medium containing a high level of potassium (K +,25 mmol/L) to another one of low potassium (K +,5 mmol/L) The cell death caused by potassium deprivation was accompanied by DNA fragmentation which was a hallmark of apoptosis Death resulted from low potassium could be prevented by caffeine in a dose dependent manner Inhibiting ryanoding sensitive Ca 2+ release including ryanodine and dantrolene did not affected the survival action, neither did L type channel blockers and NMDA receptor blocker, MK8001 Conclusion: The survival promoting effect of caffeine on cerebellar granule cells did not required increasing Ca 2+
