Abstract:Objective To explore the expression of microRNA-542-3p (miR-542-3p) in human hepatocellular carcinoma (HCC) cell lines and tissues and its effect on cell proliferation, invasion and metastasis of HCC.Methods The expression of miR-542-3p in HCC cell lines HCCLM3, Hep3B, Huh7, SMMC-7721, MHCC-97H, MHCC-97L and human normal hepatocyte LO2 was detected by RT-PCR; meanwhile, RT-PCR was also used to detect the expression level of miR-542-3p in HCC and corresponding para-carcinoma tissues. MHCC-97H and HCCLM3 cells transfected with miR-542-3p mimics were used as overexpression groups, and MHCC-97H and HCCLM3 cells transfected with empty vector were used as negative control group respectively. CCK-8 was used to detect cell proliferation, Transwell was used to detect cell invasion in vitro, Western blotting was used to detect the expression of EMT-related proteins, and immunofluorescence was used to detect the effect of miR-542-3p on EMT. Results The expressions of miR-542-3p in HCC cell lines HCCLM3, Hep3B, Huh7, SMMC-7721, MHCC-97H, MHCC-97L were lower than that in human normal hepatocyte LO2 (P<0.05). The expression of miR-542-3p in HCC tissue was lower than that in para-carcinoma tissue (P<0.05). The proliferation, invasion and metastasis in MHCC-97H and HCCLM3 overexpression groups were significantly lower than those in control group, and the fluorescence of EMT-related proteins was significantly lower than that in control group(P<0.05). Conclusion The expression of miR-542-3p is low in HCC cells. The overexpression of miR-542-3p can effectively inhibit the malignant biological behavior of HCC cells, EMT-related pathway may involve in this process. MiR-542-3p is expected to be a potential target for HCC.
