摘    要:目的从肺癌T7噬菌体展示文库筛选出能用于肺癌早期检测的分子标志群。方法亲和筛选肺癌T7噬菌体展示文库,对纯化后的富集肺癌相关肽的文库进一步行两轮ELISA检测,PCR扩增阳性噬菌体插入片段,测序后经过NCBI上的BLAST序列比对。结果筛选出13个有意义噬菌体,测序结果显示1个为未知功能的新基因,其余的均已确定与癌症相关;且13个基因中包含有三组相同的基因。结论从T7噬菌体构建的肺癌cDNA文库中筛选出的一组阳性噬菌体表达的抗原,可能是潜在的肺癌诊断分子标志群。

Screening of Molecular Markers for the Early Diagnosis of Lung Cancer from T7 Bacteriphage Display Library

Abstract:Objective To select biomarkers for early detection of lung cancer from T7 phage-peptide display library. Methods After five rounds of negative and positive biopanning, the purified phages were subjected to two further cycles of ELISA. The insert of positive phages selected by two cycles of ELISA were amplified by PCR and then sequenced. The gene alignment was blasted at NCBI to find the exact or most similar known genes. Then the relation between these genes and cancer was predicted. Results 13 positive phage clones were selected and sequenced. One of them is novel gene. Other genes have been reported that they have relation with cancer. Furthermore there are three groups of same gene. Conclusion The positive genes selected from the cDNA library constructed by T7 phage may be biomarkers for detection of lung cancer.
