摘    要:目的制备一种DNA门控的空心介孔二氧化硅纳米粒(HMSNs-DNA),观察其体外超声显像及高强度聚焦超声(HIFU)刺激触发药物释放效果。方法采用碳二亚胺法连接HMSNs和DNA制备HMSNs-DNA,检测其理化性质。体外观察HMSNs-DNA的超声成像效果。将阿霉素作为模型药物载于HMSNs-DNA纳米粒,体外运用HIFU辐照,观察其药物释放效果。结果制备的DNA门控的空心介孔二氧化硅纳米粒呈球形;随着HMSNs-DNA纳米粒浓度的增大,其超声信号强度逐渐增强;经HIFU辐照后阿霉素释放明显增强。结论成功制备DNA门控的介孔二氧化硅纳米粒,具有良好超声成像效果,并可响应HIFU刺激促发药物定位释放。

Abstract:Objective To prepare a DNA gated hollow mesoporous silica nanoparticles (HMSNs-DNA), and to observe its ultrasonic imaging in vitro and the effect of drug release by high intensity focused ultrasound (HIFU) triggering. Methods DNA gated hollow mesoporous silica nanoparticles were prepared by carbodiimide technique. Physical and chemical properties and ultrasound imaging were tested. Loading of Doxorubicin hydrochloride (DOX) in the nanoparticles, after using HIFU, the release effect was observed. Results DNA gated hollow mesoporous silica nanoparticles were spherical. The ultrasonic signal intensity enhanced with HMSNs-DNA concentration increased in vitro. After using HIFU, the release of the DOX enhanced obviously. Conclusion DNA gated hollow mesoporous silica nanoparticles which can act as ultrasonic contrast agent is successfully prepared, and the drug positioning release can be markedly improved by HIFU in vitro.
