冰冻切片技术经验探讨

目的探讨冰冻切片制作过程中的常见问题及解决方法。方法针对8 126例甲状腺、子宫、乳腺、脂肪、脑、卵巢、淋巴结、肺、皮肤、肠息肉等不同组织在冰冻切片制作过程中出现的组织脱片、组织收缩、制片困难及冰晶过多和染色时间长、染色效果不佳等问题进行改进。结果通过对取材组织的预处理,包埋剂用量的把握,冷冻时间的控制以及酒精灯和胶片的配合使用,圆满地解决了冰冻切片制作过程中的冰晶过多、、组织收缩、组织脱片等问题。结论冰冻切片中对组织取材、包埋、冷冻及染色的改进,有助于及时准确地制作出优质的冰冻切片,为明确诊断提供了良好的技术支持和条件保障。

Study on cryostat technique

Objective To explore the common problems and solutions of cryostat technique. Methods In this retrospective study,a total of 8 126 surgical fresh specimens of thyroid,uterus,breast,fat,brain,ovary,lymph nodes,lung,skin,etc. were sliced and different problems had been met,for example,tissue detachment,tissue shrinkage in different sections,creation of ice crystals,etc. The corresponding solutions had been solved. Results The improvement of surgical cutup,embedding media and freezing duration and the joint application of alcohol burner and anti-escape of slides overcoming the encountered problems. Conclusion The modified method can quickly and accurately provide good quality of sections for pathologists.
