IL-1促进子宫内膜异位症在位基质细胞增殖作用的研究

摘    要:目的:研究白细胞介素 1β(interlukin 1β,L 1β)对子宫内膜异位症(endometriosis,EMs)在位内膜基质细胞生长的影响,探讨IL 1β在EMs发病中的作用。方法:培养EMs在位内膜基质细胞作为研究EMs的体外实验模型,加入不同浓度的IL 1β后采用四唑盐比色法(MTT)观察细胞增殖情况、流式细胞计量仪(flow cytometry,FCM)检测细胞生长周期。结果:依据MTT结果所绘得的细胞生长曲线及FCM检测的细胞周期结果显示:IL 1β作用后细胞生长旺盛,G0/G1期细胞所占比例明显低于G2/M期及S期,当IL 1β的浓度为 1.0?ng/ml且作用12?h此作用最为显著。结论:IL 1β可促进EMs在位内膜基质细胞由G0/G1期进入G2/M期及S期,对子宫内膜细胞生长具有促增殖作用。

Abstract:To investigate the proliferation of endometrial stromal cells in endometriosis by interleukin 1beta(IL 1β). Methods: The eurotopic endometrial stromal cells of endometriosis cultured within 3~5 passages were treated with IL 1β in a dosage of 0 to 10.0?ng/ml, and then the cell proliferation and the cell cycle were measured by MTT and flow cytometry (FCM) respectively. Results: The endometrial stromal cells had been growing luxuriantly, and G0/G1 phase cells were few than G2/M and S phase cells. Moreover, the proliferation was the most prominent when the cells were incubated in the presence of IL 1β at 1.0?ng/ml concentration for 12 hours. Conclusions: IL 1β induces the proliferation of endometrial stromal cells of endometriosis and promotes the transformation of G0/G1 phase cells into G2/M and S phase.
