不同产地蒺藜种质鉴定和化学成分聚类分析

目的测定不同产地的蒺藜的种质遗传,和药效物质基础上的一致性。方法利用PCR产物直接测序,测定不同产地蒺藜的核糖体DNA内转录间隔区(rDNA-ITS)的基因序列;对蒺藜的化学成分进行HPLC分析,以相关系数法对色谱结果进行模糊聚类。结果测得ITS碱基序列742 bp,其中,ITS1全部序列267 bp,5.8 S全部序列167 bp,ITS2全部序列209 bp,六个产地的蒺藜样品的ITS的碱基序列完全相同;在0.90的置信水平上,六个产地的蒺藜样品聚为一类。结论不同地区的蒺藜在种质上是一致的,在药效物质基础上没有太大的差异。

Germplasm Identification and Fuzzy Clustering of Tribulus terrestris L. Collected from Different Areas of China

Objective To determine the differences between samples of Tribulus terrestris L.from different parts of China so as to see if their genotypes and their chemical substances are the same.Methods The nucleotide sequences of ITS were gained by an immediate sequencing of PCR amplifications.Using a mode of correlate coefficient,the fuzzy clustering was carried out based on the results of HPLC.Results 742 bp nucleotides of ITS in Tribulus terrestris including 267 bp of ITS1,167 bp of 5.8 S rDNA and 209 bp of ITS2 were attained,which showed no difference between samples from different parts of China.And by a confident level of 0.90,Tribulus terrestris L.from six different parts of China were clustered into one community based on their chemicals.Conclusion Different samples of Tribulus terrestris L.from different parts of China have the same genotype and there chemical substances are almost the same.
