不同辐照模式聚焦超声诱导人胰腺癌细胞凋亡的量效关系

摘    要: 目的 探讨不同辐照模式聚焦超声诱导人胰腺癌细胞凋亡的最佳辐照方案。方法 对人胰腺癌细胞系PaTu 8988t细胞悬液进行聚焦超声辐照:第1组为空白对照组,第2、3组采用脉冲辐照模式,第4、5组采用连续辐照模式,各组辐照剂量通过改变辐照时间来改变。以流式细胞仪检测辐照后24 h肿瘤细胞凋亡率及细胞死亡率。结果 超声辐照后各组的细胞悬液最高温度分别为28.00℃、(42.20±2.17)℃、(50.80±0.84)℃、(55.80±2.17)℃、(65.20±3.11)℃,各组间差异均有统计学意义(P均<0.05)。各辐照组的细胞凋亡率为(0.56±0.15)%、(1.28±0.16)%、(1.84±0.29)%、(5.74±1.15)%、(2.00±0.84)%,细胞死亡率为(3.28±0.59)%、(4.60±0.47)%、(8.64±2.69)%、(41.12±15.91)%、(49.70±8.02)%,各组间差异均有统计学意义(P均<0.05)。结论 聚焦超声辐照剂量在接近热凝固温度阈值时,采用连续辐照模式可获得较高的细胞凋亡率及细胞死亡率。
